Effects of recombinant human tumor necrosis factor-α receptor Ⅱ:Fc on NF-κB signal expressed in by synovial cells from CIA model rats
目的:探讨重组人Ⅱ型肿瘤坏死因子受体-抗体融合蛋白(rhTNFR:Fc)对胶原诱导性关节炎(CIA)模型大鼠关节滑膜细胞NF-κB信号表达的影响。方法:将40只6周龄的雌性Wistar大鼠按照随机数表法随机分为空白组、模型组、醋酸泼尼松对照组和rhTNFR:Fc治疗组,每组10只。制备CIA模型,以Western blotting法检测关节滑膜细胞NF-κB/P65、p-ⅠκBα表达的情况。结果:rhTNFR:Fc治疗组和醋酸泼尼松对照组可下调CIA大鼠滑膜细胞NF-κB/P65、p-ⅠκBα的表达,与模型组比较均有显著性差异(P<0.05)。结论:rhTNFR:Fc可通过降低CIA大鼠关节滑膜细胞NF-κB/P65、p-ⅠκBα蛋白表达以达到缓解炎症的作用。
OBJECTIVE To discuss how recombinant human tumor necrosis factor-α receptor Ⅱ:Fc(rhTNFR:Fc) influences expression of NF-κB signal in synovial cells from CIA model rats and inflammatory factors.METHODS Forty six-week-old female Wistar rats were randomly divided into 4 groups including blank control group(n=10), model control group(n=10), treatment group with rhTNFR:Fc(n=10) and prednisone acetate control group(n=10). Through establishing CIA model, detecting expression of NF-κB/P65 and p-ⅠκBα in synovial cells with Western blotting. RESULTS Compared with model control group, rhTNFR:Fc and prednisone acetate could visibly reduce expression of NF-κB/P65 and p-ⅠκBα in synovial cells from CIA model rats(P<0.05).CONCLUSION rhTNFR:Fc can alleviate inflammation through remarkably reducing expression of NF-κB/P65 and p-ⅠκBα in synovial cells from CIA model rats.
